STABLE PICKERING EMULSION STABILIZED BY ACETYLATED CELLULOSE NANOCRYSTALS, METHOD FOR PRODUCTION AND USE THEREOF

SUBSTANCE: invention relates to pharmaceutical industry, namely to a stable Pickering emulsion. Stable Pickering emulsion, which is a heterogeneous system consisting of two immiscible liquids, in which one liquid is uniformly distributed throughout the volume of the other liquid in the form of oil drops stabilized by nanocrystalline cellulose particles, characterized by that the first liquid in the form of drops is Vaseline or olive oil, and the second is a hydrosol with content of acylated nanocrystalline cellulose (ACNCC) in amount of 0.2–1.6 wt. % with zeta potential of particles minus 36–40 mV, particle size along length from 135 nm to 205 nm and cross section - from 6 nm to 10 nm, wherein in an emulsion consisting of a dispersion aqueous medium and oil drops, the components are distributed at the following volume ratio: hydrosol - 70 %; vaseline or olive oil - 30 %, wherein the surface of the oil drops in the emulsion has a negative zeta potential, the zeta potential of the drop for the emulsion with Vaseline oil minus 6.9, minus 7.5, minus 9.6, minus 10.3 or minus 12.5, for emulsion with olive oil - minus 10.9, minus 11.9, minus 17.7, minus 18.5 or minus 19.9. Method of producing stable Pickering emulsions, comprising emulsifying two immiscible liquids until one liquid is evenly distributed throughout the volume of the other liquid to form oil drops stabilized in the volume, characterized by that emulsification is carried out with ultrasound until the oil is uniformly distributed throughout the volume of the hydrosol in the form of oily drops using an ultrasonic generator IL-10-0.1 for 180 seconds with frequency of 22 kHz, stabilization of the surface of oil drops is carried out with particles of ACNCC with a zeta potential of particles of minus 36–40 mV, with length of 135 nm to 205 nm and a cross section of 6 nm to 10 nm, wherein during emulsification of vaseline or olive oil and hydrosol containing ACNCC in amount of 0.2–1.6 wt. %, obtaining a stable shell on the surface of drops with a negative zeta potential: for an emulsion with Vaseline oil minus 6.9, minus 7.5, minus 9.6, minus 10.3 or minus 12.5, for emulsion with olive oil - minus 10.9, minus 11.9, minus 14.0, minus 17.7, minus 18.5 or minus 19.9. Disclosed is the use of an emulsion which is stable under conditions of the stomach as a target delivery system of lipophilic drugs when administered orally.
EFFECT: emulsion described above is a gastric-stable lipophilic drug delivery system; the method for preparing the emulsion is effective and simple; it enables to form a stable coating of emulsified drops.
