The invention relates to a detection method in the field of plant disease resistance molecule breeding, particularly to a multiplex PCR method for simultaneously detecting tomato Ty-1 gent, tomato Ty-3 gene and tomato Mi gene, and applications thereof. According to the present invention, by using the multiplex PCR method, the tomato disease resistance germplasm resources can be screened and identified; by using four pairs of primers closely linked to the tomato Ty-1 gent, the tomato Ty-3 gene and the tomato Mi gene, the genotypes of the three tomato genes can be efficiently and accurately detected through the one PCR reaction and the gel electrophoresis; the results of the multiplex PCR reaction gel electrophoresis are consistent with the results of the single gene PCR reaction gel electrophoresis, such that the results show that the method can be effectively utilized; and by using the method, the detection time and the detection cost of the three disease resistance genes such as the tomato Ty-1 gent, the tomato Ty-3 gene and the tomato Mi gene can be substantially saved, and the detection results can effectively assist the tomato disease resistance breeding work.
