The invention relates to a cultivation method and application of tomato grafted seedlings. The cultivation method comprises the following steps of (1) respectively disinfecting and accelerating germination of root-knot nematode-resistant pepper seeds and tomato seeds, sowing by stages, and cultivating root-knot nematode-resistant pepper seedlings and tomato seedlings; (2) selecting the root-knot nematode-resistant pepper seedlings as stocks and the tomato seedlings as scions when the root-knot nematode-resistant pepper seedlings reach a period of 4-5 true leaves and the tomato seedlings reach a period of 3 leaves and 1 core, processing the middle of the section of a tender stem of each root-knot nematode-resistant pepper seedling to form a cleaved notch, processing a cotyledonary node of each tomato seedling to form a tip, and performing grafting, specifically, inserting the tips of the tomato seedlings into the cleaved notches of the root-knot nematode-resistant pepper seedlings to be aligned and tightly attached, using grafting clamps for clamping the joints, and completing grafting; and (3) after completing grafting, spraying tomato grafted seedlings, putting the tomato grafted seedlings into a plastic arched shed for moisturizing, culturing for 2-3 weeks after grafting, transplanting and planting, and carrying out production management. The tomato grafted seedlings cultivated by the cultivation method disclosed by the invention has a root-knot nematode prevention and control effect of 100%.
