The invention discloses a process for preparing melatonin from tomato raw materials via combined enzymatic hydrolysis extraction, belonging to the technical field of melatonin preparation. The process comprises: S1, preparing materials including 100–680 parts of tomatoes, 10–120 parts of black ginger, 20–120 parts of rice germ polyamines, 10–100 parts of pectinase, 5–50 parts of cellulase, 10–30 parts of a compound enzyme preparation, 3–30 parts of an extraction solvent, 2–55 parts of a pH regulator, 1–40 parts of a stabilizer, 10–110 parts of gel chromatography packing, and 100–2000 parts of deionized water; and S2, pretreating raw materials, wherein tomatoes are processed into slurry and black ginger is processed into powder (80–120 mesh).The process improves product stability and storage life, and is green, safe, simple, controllable, suitable for industrial production, and economical, providing an efficient route for high-purity melatonin preparation.
