Host cells, such as immune cells, including the molecules, antibodies, fusion proteins, CARs, nucleic acids encoding them, are also provided. In some forms, the cells are CAR immune cells. In some forms, the CAR immune cells maintain the ability to kill ENPP3+ cancer cells after multiple stimulations. Pharmaceutical compositions including the antibodies, CARs or T cells are also provided. In some forms, the molecule or antibody includes drugs conjugated thereto, has antibody-dependent cell-mediated cytotoxicity (ADCC) activity, complement-dependent cytotoxicity activity (CDC), and/or is a bi-specific engager optionally a T cell engager or NK cell engager. Methods of using the described molecules to treat ENPP3+ cancers are also described.
