The invention provides a method to increase the yield of recombinant milk casein proteins in transgenic soybean by co-expressing those caseins together with a combination of protease inhibitors — specifically a cysteine protease inhibitor, a serine protease inhibitor, and a metalloprotease inhibitor — that are targeted to the same subcellular compartment as the caseins. The core problem being solved is that endogenous soybean proteases degrade the foreign casein proteins before they can accumulate to commercially useful levels. The solution is to escort the caseins through their entire secretory lifespan with inhibitors that collectively neutralize the major protease classes responsible for that degradation. The most directly demonstrated embodiment uses NbPot1 (serine) and NbPR4/CaPR4c (cysteine) co-expressed with PSV-targeted caseins in soybean zygotic embryos. The practical goal is a sustainable, plant-based production platform for milk proteins with applications in dairy alternatives, infant formula, and functional food ingredients.
