The method may comprise coupling a nucleic acid adapter to an end of a double stranded nucleic acid molecule and/or coupling an additional nucleic acid adapter to an additional end of the double stranded nucleic acid molecule, to form a chimeric nucleic acid molecule. The nucleic acid adapter may be a double stranded nucleic acid molecule. At least a portion of a first strand of the nucleic acid adapter may be conjugated to at least a portion of a second strand of the nucleic acid adapter, to form a conjugated adapter comprising a single stranded portion. The conjugated adapter may be coupled to both strands of the end of the double stranded nucleic acid molecule.
