WIPO "tomato"
A chimeric protein comprising an amino acid sequence for an icosahedral virus capsid protein and a heterologous peptide. The heterologous peptide is operably linked at a position which is (i) located on the surface, (ii) not be localised within a beta-sheet or alpha-helix, (iii) not located at a position required for the formation of pentamer and hexamer substructures, and (iv) where the side chains are not facing inwards. The icosahedral virus can be Tomato Aspermy Virus (TASV). The heterologous peptide can be inserted after arginine 46. Threonine 47 can be deleted. The heterologous protein can be Im7, Barstar, or SpyCatcher. The heterologous peptide can be from the IL-6 family of cytokines such as human or canine IL-31, human IL-13. A further aspect is a virus-like-particle (VLP) comprising the chimeric protein. A further aspect is a modified VLP comprising a second chimeric protein bound to the heterologous peptide. A further aspect is a polynucleotide or vector encoding the chimeric protein and cells and cell cultures. The chimeric protein or VLP can be used as a medicament for the prevention and treatment of inflammatory diseases, cancer, neurodegenerative, metabolic, cardiovascular, renal, gastrointestinal, respiratory, dermatological, endocrine, rheumatological or autoimmune diseases.
