Genetically engineered rice is processed and crushed, and is then mixed with an extraction buffer, and extraction and filtering are performed to obtain a crude extract containing recombinant reteplase; and separation and purification steps sequentially comprise: 1) performing cation exchange chromatography on the crude extract containing the recombinant reteplase to obtain a primary product I; 2) performing hydrophobic chromatography on the primary product I to obtain an intermediate product II; and 3) performing benzamidine affinity chromatography on the intermediate product II to obtain purified recombinant reteplase.
