The preparation method comprises the following steps: (1) linking bee PGRP-SA gene segments to an expression vector, and converting the bee PGRP-SA gene segments into escherichia coli for expression, and collecting inclusion body proteins obtained through expression; (2) washing the inclusion body proteins, adding a protein denaturant for dissolving, and carrying out renaturation by virtue of renaturation liquid; and (3) concentrating the proteins, and carrying out elution and purification by virtue of a molecular sieve column and an anion exchange column. According to the preparation method, the expression conditions of bee PGRP-SA proteins are optimized, a formula of important reagents in each process is optimized, and chromatographic renaturation is combined and adopted, so that the method is simple and rapid, the operation process is mild, the protein damage is low, and the active protein with high purity is obtained.
