Analysis of aroma and key differential volatile compounds in mainstream smoke of cigars using HS-SPME-GC-MS-O and HS-GC-IMS

Online ahead of print.ABSTRACTThis study sought to elucidate the differences in the composition of mainstream smoke components among various hand-rolled cigars. The volatile profiles of cigar smoke were analyzed using headspace solid-phase microextraction in conjunction with HS-SPME-GC-MS-O and HS-GC-IMS techniques. Through the application of chemometric analysis, key aroma-active compounds were identified based on relative odor activity value (ROAV) and variable importance in projection (VIP) metrics. The findings revealed that only two compounds were consistently detected by both analytical methods. Specifically, 124 compounds, including esters, alcohols, and aldehydes, were identified using HS-GC-IMS, while 60 compounds were identified using HS-SPME-GC-MS-O. ROAV analysis identified 10 key aroma-active compounds with ROAV ≥ 1. Among these, based on variable importance in projection (VIP > 1) and sensory verification, 5 compounds were further recognized as the core sensory contributors, including 3-methylindole, p-cresol, 4-hydroxystyrene, 3-ethyl-4-methyl-1H-pyrrole-2,5-dione, and 2,3'-bipyridyl. The Partial Least Squares Discriminant Analysis (PLS-DA) model effectively differentiated the five samples, with differential compounds having VIP scores greater than 1, including alcohols, phenols, heterocyclic compounds, and aldehydes. The sensory radar chart revealed distinct aromatic profiles for each sample: Sample T1 was characterized by woody, nutty, and roasted notes; Sample T2 exhibited a pronounced honey-like sweetness and a beany aroma; Sample T3 displayed a balanced profile of honey-like sweetness and beany notes; Sample T4 was dominated by woody and floral aromas; and Sample T5 featured prominent roasted and fruity notes. This study illustrates that the complementary application of HS-SPME-GC-MS-O and HS-GC-IMS offers an effective methodology for elucidating the complex aroma profile of cigar mainstream smoke.
