According to the invention, genotypes of V-type sucrose esters in three different types of tobacco sucrose esters are distinguished for the first time, and a codominant SSR marker closely linked with the V-type tobacco sucrose ester gene qBMVSE499 is provided. The marker can be used for qualitatively or quantitatively detecting the V-type tobacco sucrose ester and the genotype state of the V-type tobacco sucrose ester in a plant to be detected. Compared with a method in the prior art, the method has the advantages of higher detection accuracy, stability, reliability, simplicity and convenience in operation, low cost and no limitation on detection opportunity.
