The method comprises the following steps that dibble seeding is carried out on M2-generation seeds of tobacco Yunyan 87 to floating seedling raising plates; when two main leaves grow from each Yunyan 87 control material, seedlings are pulled out and cleaned, root systemconditions are compared, and screened single plants are transferred into small flowerpots to be cultured for 20 days; the tobacco plants are transplanted into large flowerpots after 30 days; and thenfield planting is carried out, fertilization is carried out, selfing seed harvesting is carried out, and the root system conditions of the tobacco mutants obtained through screening are determined again in the second year. According to the method, the tobacco EMS mutation populations are screened on a large scale, the root systems of the mutant tobacco materials are observed, the tobacco mutant materials with the root systems stronger than those of wild type tobacco mutant materials are obtained by screening, and a good foundation is laid for tobacco breeding work in the future.
