The invention discloses a method for detecting the content of proanthocyanidins in tobacco leaves, which can be used for simultaneously detecting catechin, epicatechin, proanthocyanidins B1 and proanthocyanidins B2 in the proanthocyanidins in the tobacco leaves, and specifically comprises the following steps: pretreating a sample, extracting, purifying and detecting the proanthocyanidins. According to the method, the procyanidine in the tobacco sample is extracted through a rapid, simple and reliable extraction method, the matrix effect is removed by adding the Cardon dispersion adsorbent, and the residual quantity of the procyanidine in the tobacco leaves is determined by adopting the high-selectivity and high-sensitivity ultra-high performance liquid chromatography; and compared with the prior art, the detection method disclosed by the invention has the characteristics of simplicity and convenience in sample treatment and higher detection sensitivity and accuracy, and can completely meet the detection requirements of the procyanidine.
