The invention discloses a method for creating tobacco interspecific allopolyploidy new germplasm by using horizontal genome transfer, and belongs to the technical field of tobacco polyploidy breeding. The method comprises the steps of sterile seedling cultivation, in-vitro grafting, grafting combination part callus induction, adventitious bud induction and rooting culture, regenerated plant transplanting, regenerated plant phenotype screening and identification, ploidy identification and the like. According to the method, nicotiana tobacco and aztec tobacco are used as grafting parent materials, and a test tube in-vitro grafting technology is used for achieving tobacco genome horizontal transfer; and with adoption of an artificial tissue culture technology, grafting joint callus induction and asexual strain regeneration of the two grafting parents are realized, sexual breeding obstacles between two related tobacco species are overcome, and a fertile tobacco allopolyploidy plant is formed.
