The invention discloses a method and device for quickly detecting tobacco maturity based on tobacco differential metabolite content, and the method comprises the steps: carrying out the non-targeted metabolite detection and analysis of tobacco metabolite through LC-MS, carrying out the data preprocessing and statistical analysis through an XCMS online platform, screening out the tobacco differential metabolite, constructing a molecular structural formula library, and determining the nature of the screened differential metabolite in combination with a mass spectrum prediction method in MS-FINDER software, and screening one or more differential metabolites for judging the maturity of the tobacco leaves; and judging the maturity of the tobacco leaves by detecting and judging the ion flow intensity of the differential metabolite of the maturity of the tobacco leaves. According to the device, a spectral data acquisition unit is used for acquiring an infrared light absorption value of a tobacco leaf sample to be detected, the ion flow intensity of the differential metabolite used for judging the maturity of the tobacco leaves is predicted through a prediction model, and finally the maturity of the tobacco leaves to be detected is judged according to a judgment model. According to the method and the device, the maturity of the tobacco leaves can be quickly and accurately judged, and a guarantee is provided for subsequent tobacco leaf curing.
