The construction method comprises the following steps of: cloning a glurmaceren-like synthetase C coding gene to an expression vector by adopting a recombinant DNA technology, and transforming a host to obtain a recombinant strain of high-yield glurmaceren-like synthetase C; and performing directed evolution on amino acid residues on a loop structure around an active site E474 of the glurmaceren-like synthetase C, and screening to obtain a high-activity glurmaceren-like synthetase C mutant. The invention further provides application of the method to preparation of a product containing cemeterol. By adopting the high-activity glurmaceren-like synthetase C mutant and the method obtained by the invention, cemeterol can be produced efficiently under catalysis, and application of cemeterol to the fields of agriculture,tobacco and the like can be promoted.
