The invention provides an ethanol preparation method capable of reducing tobacco stem pretreatment product inhibition influence. The method comprises the following steps: (1) cleaning waste tobacco stems, drying in the sun, crushing with a crusher, and sieving; (2) treating the crushed tobacco stems with a dilute sulfuric acid solution; (3) carrying out solid-liquid separation after acid pretreatment, drying the solid part without washing, grinding into powder, and sieving; (4) preparing a fermentation culture medium by taking the pretreated tobacco stem powder as a carbon source, and adjusting the pH value to 5.4-5.6; and (5) adding cellulase into the fermentation medium, adding sodium borohydride, inoculating a 4-6% yeast seed solution, and carrying out ethanol fermentation in a shaking table at 28-32 DEG C for 40-54 hours. The method has the advantages of low raw material cost, no need of a washing process, environmental protection, production cost saving, avoiding of the generation of a large amount of sewage, solving of the environmental pollution problem of the raw materials, realization of turning waste of the tobacco stems into wealth, and extremely high feasibility, and is an efficient and environment-friendly bioethanol fermentation process.
