Verification method of Meloidogyne in tobaccos

The invention discloses a verification method of Meloidogyne in tobaccos. The verification method integrates morphological and molecular characteristics, and comprises the following steps: 1) morphological identification; 2) DNA extraction; 3) LSU 28S rDNA D2-D3 sequence amplification: carrying out PCR (polymerase chain reaction) amplification on the extracted DNA in the step 2) by using universal primers RK28SF/MR for Meloidogyne, detecting, preliminarily verifying the wireworm to be detected is Meloidogyne if a 614bp sequence is obtained, and establishing a molecularly-phylogenetic tree; 4) according to the established molecularly-phylogenetic tree, determining whether the wireworm to be detected is Meloidogyne through phylogenetic development relation analysis; and 5) SCAR-PCR detection on sequence characteristic amplification region. The method provides a powerful technical system for efficient accurate verification of tobacco Meloidogyne varieties, and provides favorable foundation for mastering the distribution and damage extent of Meloidogyne in the tobacco planting region.
