The invention discloses a transcription factor NtMYB44b capable of improving the drought resistance of tobacco and a fixed-point mutation method and application of the transcription factor. A nucleotide sequence of the transcription factor NtMYB44b capable of improving the drought resistance of the tobacco is shown in SEQ ID NO:1. An amino acid sequence of coding protein is shown in SEQ ID NO:2; by adopting a CRISPR/Cas9 technology, one sgRNA target site is designed and located at a distance stopping codon, and by transforming the tobacco, mutation of the gene at a target site is caused; through selfing and seed harvesting, NtMYB44b gene fixed-site homozygous mutation offspring are obtained through screening. It is proved through a physiological experiment that the water loss rate of mutation tobacco leaves is obviously lower than that of a wild-type check plant. It is indicated through a water overlying experiment after drought stress that the survival rate of a mutant plant is obviously higher than that of the wild-type plant. It is indicated that by using the CRISPR/Cas9 technology, compared with the wild-type tobacco, a NtMYB44b gene homozygous mutation tobacco material manufactured for the tail of the NtMYB44b gene has the higher drought resistance capacity, and the gene resources and technical support are provided for drought resistance breeding of the tobacco.
