The invention discloses a trace maleic hydrazide separation method, which comprises steps such as pretreatment and extraction-separation, and specifically comprises: drying raw materials, crushing, and screening; placing into an extraction tank, and extracting with a supercritical CO2 fluid to obtain an extraction liquid; carrying out evaporation concentration on the extraction liquid to obtain an extraction concentration liquid, adding methanol to the extraction concentration liquid, uniformly shaking, and filtering with an organic filtration membrane to obtain an extraction purification liquid, wherein a mass volume ratio of the methanol to the raw material is 1:1-5; and detecting the maleic hydrazide content in the extraction purification liquid by using high performance liquid chromatography. According to the present invention, the supercritical carbon dioxide fluid and the special auxiliary agent are adopted as the extractant to extract the trace maleic hydrazide in the tobacco leaf, and the obtained extraction liquid is subjected to the concentration and purification treatment so as to be used for the high performance liquid chromatography analysis and detection; and the method has advantages of simple operation process, high extraction rate, less organic reagent consumption, quick pre-treatment, high recovery rate, good stability and no damage to the determined sample, and can be used as the special determination method for the trace maleic hydrazide in tobacco and tobacco products.
