The tobacco seedling-raising substrate comprises, by weight, 40-60 parts of coconut coir, 5-15 parts of grass carbon, 20-40 parts of perlite and 5-15 parts of vermiculite. The preparation method comprises the steps of pretreatment and preparation, and comprises the specific steps of weighing the coconut coir, grass carbon, perlite and vermiculite according to the ratio of the formula, mixing the coconut coir, grass carbon, perlite and vermiculite to be uniform, and the target product is obtained. According to the application, the tobacco seedling-raising substrate is applied to the preparation of a nursery site which is high in survival seedling rate and can protect a wetland ecological system. According to the preparation method, the coconut by-products or waste, the perlite with stable physical and chemical properties, the vermiculite and the grass carbon are used as main materials to prepare the tobacco seedling-raising substrate; on the basis that the tobacco seedlings come out evenly and the survival seedling rate is high, the tobacco seedling-raising substrate reduces the usage amount of the grass carbon, protects the wetland ecological system, takes full advantage of renewable resources, and plays a greater role in promoting the sustainable development of tobaccos.
