The tobacco seedling culture substrate comprises the following raw materials in parts by weight: 40 to 60 parts of coconut husk, 2 to 8 parts of lignite, 20 to 40 parts of perlite and 5 to 15 parts of vermiculite. The preparation method comprises the pretreatment and preparation steps, and specifically comprises the following steps of crushing the raw lignite for standby; weighing the coconut husk, lignite, perlite and vermiculite according to formula, and uniformly mixing, so as to obtain a target matter. The application is characterized in that the tobacco seedling culture substrate is applied to prepare seedling culture pots with high seedling survival rate and capable of protecting a wetland ecosystem. The tobacco seedling culture substrate has the advantages that the coconut byproduct or waste, the perlite with stable physical and chemical property, and the vermiculite are used as the raw materials and are prepared into the tobacco seedling culture substrate; the emergence of tobacco seedling is regular, the seedling survival rate is high, and the destroy of the wetland ecosystem is avoided; the regenerated resources can be fully utilized, and the larger promoting function on the sustained development of tobacco is realized.
