The invention discloses a tobacco chloride ion absorbing gene NtSLAC2 and a cloning method and application thereof. The nucleotide sequence of the tobacco chloride ion absorbing gene NtSLAC2 is as shown in SEQ ID: No. 1, and a coded amino acid sequence is as shown in SEQ ID: No. 2. The invention further discloses a cloning method of the tobacco chloride ion absorbing gene NtSLAC2. The cloning method comprises the following specific steps: extracting tobacco RNA, and carrying out reverse transcription to obtain a first chain cDNA; and designing and synthesizing specific primers by taking the first chain cDNA obtained by reverse transcription as a template according to the gene NtSLAC2, and carrying out PCR amplification, recycling and purifying a PCR amplification product, and sequencing the PCR amplification product. In the invention, expression of a tobacco endogenous gene NtSLAC2 is inhibited in tobacco plants, the content of chloride ions of tobacco leaves can be reduced obviously,the application prospect in the field of low-chlorine-ion-content breeding of plants is wide, and the potential economic benefits of the tobacco chloride ion absorbing gene NtSLAC2 are huge.
