The invention discloses tobacco axillary bud growth regulating gene NtMOC1 and a cloning method and application thereof. The cloning method of the tobacco axillary bud growth regulating gene NtMOC1 includes steps: A, determining a NtMOC1 gene sequence; B, extracting tobacco RNA to obtain first chain cDNA through reverse transcription; C, designing according to the NtMOC1 gene sequence to synthesize a specific primer, and taking cDNA as a template to perform PCR (polymerase chain reaction) amplification; D, recovering and purifying a PCR product; E, connecting the purified product with a carrier to transform competent cells; F, screening positive clones, and subjecting the positive clones to PCR amplification and sequencing. By regulation of expression of the tobacco axillary bud growth regulating gene NtMOC1, formation of tobacco axillary buds can be inhibited after topping, and accordingly consumption of tobacco bud inhibitors can be reduced hopefully to improve tobacco quality, and tobacco production labor intensity, manual and chemical agent cost and environmental pollution can be reduced.
