The invention discloses a tobacco auxiliary bud growth regulation and control gene NtMAX3-2 as well as a cloning method and application thereof. The nucleotide sequence of the tobacco auxiliary bud growth regulation and control gene NtMAX3-2 is shown in SEQ ID No.1, and an amino acid sequence encoded by the gene is shown in SEQ ID No.1. The invention further discloses a cloning method of the tobacco auxiliary bud growth regulation and control gene NtMAX3-2. The cloning method specifically comprises the following steps: A, confirming a NtMAX3-2 gene sequence; B, extracting tobacco RNA (Ribonucleic Acid), and performing inverse transcription so as to obtain first-chain cDNA (Complementary Deoxyribonucleic Acid); C, according to the NtMAX3-2 gene sequence, designing and synthesizing a specific primer, and performing PCR (Polymerase Chain Reaction) amplification by taking cDNA as a template; D, recycling and purifying a PCR product; E, connecting the purified product with a carrier, and converting competent cells; F, screening positive cloning, and performing PCR amplification and sequencing on the positive cloning. By regulating and controlling expression of the tobacco auxiliary bud growth regulation and control gene NtMAX3-2, formation of tobacco auxiliary buds after top pinching is inhibited, so that use of a tobacco bud inhibitor is reduced, and a gene resource is provided for producing green high-quality tobacco leaves and reducing labor cost.
