The invention discloses a quantitative analysis method of maleic hydrazide and glycosides of maleic hydrazide in tobacco leaves based on liquid chromatography-tandem mass spectrometry. The quantitative analysis method comprises the following steps: (1) sample pretreatment: accurately weighing tobacco leaf sample powder, putting in a centrifugal tube, adding methanol, methyl tert-butyl ether and water as extractants in sequence, performing ultrasound treatment and centrifugation, tanking subnatant, adding an internal standard solution, performing centrifugal concentration to remove a solvent, adding acetonitrile to redissolve, and transferring a redissolving solution into a liquid chromatography sample injection vial for analysis; (2) drawing a standard curve; (3) sample analysis: performing liquid chromatography-tandem mass spectrometry analysis on the to-be-analyzed sample treated in the step (1), inputting obtained chromatography mass spectrometry peak relative area of the maleic hydrazide and the glycosides of the maleic hydrazide into a calibration curve equation, and calculating to obtain corresponding substance concentration. The quantitative analysis method creatively detects the content of the maleic hydrazide in a free state and the maleic hydrazide glycoside in a combined state in the tobacco leaves, thus greatly improving the accuracy of analysis results, and the method is simple and highly efficient.
