The invention discloses a quantitative analysis method for glyphosate residues in tobaccos based on gas chromatography-tandem mass spectrum. A tobacco sample is weighed and placed into a centrifugal tube with a cover, high-purity water is taken, ultrasonic-assisted extraction is carried out, centrifugation is performed, supernate is taken and subjected to concentration by centrifuging so as to remove solvents, then pyridine and a derivatization reagent MSTFA are sequentially added, water-bath derivatization is carried out for 30 min, a product is placed at the normal temperature for 2 h afterderivatization, and a to-be-detected sample is obtained; a glyphosate mixed standard solution is prepared, and a calibration curve equation and a linear correlation coefficient are obtained through gas chromatography-tandem mass spectrum analysis; and the to-be-detected sample is subjected to gas chromatography-tandem mass spectrum analysis to obtain the gas chromatography-mass spectrometry peak area of glyphosate in the to-be-detected sample, and the peak area is input to the calibration curve equation for calculation to obtain corresponding material concentration. Analysis recovery of glyphosate in tobaccos is 95.1-103.5%, the detection lower limit is 4 mg/kg, the quantitative lower limit is 10 mg/kg, the intra-day repeatability is 3.53%, and the inter-day repeatability is 5.16%.
