The invention discloses an NteIF4E-1 gene mutant for generating PVY resistance and an application thereof. Relative to an NteIF4E-1 gene with a nucleotide sequence shown as SEQ ID NO.1, the NteIF4E-1 gene mutant for generating PVY resistance is characterized in that G on No.159 locus in NteIF4E-1 gene sequence contained in the NteIF4E-1 gene mutant for generating PVY resistance is replaced by A and the point mutation occurs. The nucleotide sequence of the NteIF4E-1 gene mutant for generating PVY resistance is shown as SEQ ID NO.3. The application is the application of the NteIF4E-1 gene mutant for generating PVY resistance in acquiring the tobacco plant with PVY resistance. The invention is created through EMS mutant of flue-cured tobacco Yunyan 87. A Tilling technique is utilized to screen the NteIF4E-1 gene mutant strain, perform sequencing verification on objective mutant in mutation material and analyze the PVY resistance of the mutant material with homozygosis termination mutation of NteIF4E-1 gene. The flue-cured tobacco material with termination mutation of NteIF4E-1 gene acquired according to the method can resist against the PVY virus.
