The method comprises the following steps: carrying out knockout (Knockout, KO) on an eIFiso4E-S gene contained in the tobacco plant to obtain an eifiso4e-sKO mutant, and polymerizing the mutuant and va or eif14e1KO gene to achieve the resistance of the tobacco to the ChiVMV. The methodhas great application prospect for cultivating the anti-ChiVMV tobacco.
