The method comprises the following steps: fixing the tobacco seeds with the fixed quantity on a dough piece plane made of edible flour; cutting the seeds in batches by adopting a freezing microtome; washing the cut seeds by using water and placing the seeds into a 0.01%-0.1% of iodonitrotetrazolium purple solution; dyeing in a dark environment for 2-5 hours and observing and identifying under a stereoscopic microscope, wherein the percentage of the seeds with embryos and endosperms which are totally dyed to be red in the total quantity of the detection seeds is the vitality of the tobacco seeds. According to the method, the determination time of the vitality of the tobacco seeds is greatly shortened to be 2.2-5.2 hours from 68-70 hours; the detection speed is fast and the result is accurate and reliable; the method for cutting the seeds is low in labor intensity and high in working efficiency; the working time is shortened greatly. The method for rapidly determining the vitality of the tobacco seeds has a good application prospect.
