The method comprises the following steps: firstly, spraying 65% zineb wettable powder to a cultivated tobacco plant, wherein the dosage of each mu of land is 150 g; taking samples of the 10th, the 11th and the 12th leaves of each tobacco plant 2 hours after zineb spraying and at the 1st day, the 3rd day, the 5th day, the 7th day, the 10th day, the 14th day, the 21st day, the 28th day, the 35th day and the 42nd day, testing the contents of the zineb residues, and drawing a variation curve; taking samples at the 42nd day after zineb spraying according to the variation curve, wherein the sampled tobacco leaves can be used after being baked, crushed, screened and uniformly mixed, and the content of zineb in the flue-cured tobacco standard sample natural substrate positive material is 5.40+/-0.34 mg/kg. By adopting the method, a natural substrate standard sample which is accordant with a real detection sample and is applied to analysis and quality control on zineb residues in flue-cured tobacco can be prevented, the accordance of extraction rates of a quality control sample and a detection sample under identical conditions is ensured, and the prepared natural substrate standard sample is uniform, stable and convenient to store and use.
