A method for evaluating root-knot nematode resistance of tobacco varieties by potted seedlings with diseases relates to the technical field of tobacco disease-resistant breeding, and comprises the following steps: inoculating root-knot nematode to root systems of seedlings to form seedlings with diseases, transplanting the seedlings with diseases to a draining and breathable matrix, and forming typical diseases on the root systems of tobacco plants under the conditions of proper temperature, humidity and soil permeability, pouring into pots, taking roots, cleaning, investigating diseases, and evaluating disease resistance. The method is carried out under controllable environmental conditions, quantitative single-plant seedling inoculation is carried out, nematodes are in close contact with plant root systems, the time for different plant root systems of the same variety to infect nematodes is concentrated, the disease development speed is high, the severity is more uniform and consistent, the accuracy and stability of tobacco root knot nematode disease resistance identification are improved, and the method is suitable for large-scale popularization and application. The defects that traditional potting disease soil is long in disease attack time and uneven in disease attack are overcome. Meanwhile, compared with the prior art, the method has the advantages that the identification period is shortened by 15-20 days, the workload is greatly reduced, and the method is easier to operate and popularize.
