The method comprises the steps of: heating, sterilizing, drying and cooling balls to the room temperature for standby application; dripping a bacterium suspension liquid onto a culture medium panel, then adding the balls, covering a petri dish cover of the culture medium panel, and shaking the culture medium panel until the balls uniformly apply the bacterium suspension liquid on the surface of the culture medium panel; and opening the petri dish cover to pour the balls out, and sterilizing and cleaning the balls for repeated use. According to the method, the balls serve as application tools, the culture medium panel is shaken, and the balls are adhered with the bacterium suspension liquid to freely roll on the surface of the culture medium panel, so that the bacterium suspension liquid is uniformly applied. A rolling application mode is adopted, the surface of the culture medium panel is not easy to scratch, the bacterium suspension liquid is uniformly applied, the application efficiency is high and quick; most importantly, ball application is utilized, opening or semi-opening operation does not need, the contamination degree is decreased; and moreover, the balls can be sterilized and stored in a centralized manner, and the time for preparing the application tools is shortened.
