The invention discloses a method for acquiring a natural substrate positive material of an imidacloprid-remained flue-cured tobacco standard sample. The method comprises the following steps: spraying imidacloprid water dispersible granules having a concentration of 70 percent to cultured tobacco plants with an application dosage of 3g each Mu; sampling the 10th, 11th and 12th tobacco leaves of each tobacco plant in2 hours, 1 day, 3 days, 5 days, 7 days, 10 days, 14 days, 21 days, 28 days, 35 days and 42 days after application, and detecting the remaining content of imidacloprid, and making a variation curve; sampling on the 5th, 10th and 14th day after application according to the variation curve, baking and grinding the picked tobacco leaves, screening and uniformly mixing. The content of imidacloprid in the natural substrate positive material of a flue-cured tobacco standard sample is 3.30+/-0.22mg/kg. The method can be used for preparing a natural substrate standard sample which is consistent with a real detection sample and is used for analysis and quality control of imidacloprid remained in flue-cured tobaccos, so that the extraction rate uniformity of a quality control sample and a detection sample under the same condition can be ensured. The prepared natural substrate standard sample is uniform and stable, and is convenient to store and use.
