Liquid chromatogram-high resolution mass spectrum (LC-MS) based quantitative analysis method of tobacco leaf maleic hydrazide and glucoside thereof

The invention discloses a liquid chromatogram-high resolution mass spectrum (LC-MS) based quantitative analysis method of tobacco leaf maleic hydrazide and glucoside thereof. The analysis method comprises the following steps: (1) sample pretreatment: precisely weighing a sample of tobacco leaf powder, adding an extracting agent composed of methanol, methyl tert-butyl ether, and water, carrying outan ultrasonic treatment and centrifugation, saving lower layer supernate, adding an internal standard solution, carrying out centrifugal concentration to remove the solvent, adding acetonitrile to carry out re-dissolution, transferring the solution to a LC sample bottle, and storing the LC sample bottle for later analysis; (2) drawing a standard curve; (3) sample analysis: subjecting the sample obtained in the step (1) to LC-MS analysis, inputting obtained relative areas of LC-MS peaks of tobacco leaf maleic hydrazide and glucoside thereof into the standard curve equation, and calculating thecorresponding concentrations of tobacco leaf maleic hydrazide and glucoside thereof. The provided method creatively detects the contents of free state maleic hydrazide and combined state maleic hydrazide glucoside in tobacco leaves, the accuracy of analysis results is greatly improved, the method is convenient, and the efficiency is high.
