The seed multiplication method includes the steps of carrying out seed cleaning on a tobacco genetic bank, listing various tobacco genetic resources with seed storage amount smaller than 3g, screening out the genetic resources which are high in death rate because of infection of root stock diseases like blackleg and bacterial wilt in seed multiplication processes over the years, adopting varieties resistant to the tobacco root stock diseases as stocks, adopting susceptible genetic resources as cions for grafting, receiving seeds of cion varieties and storing the seeds. The grafting and seed multiplication method is simple, capital and manual investment is little, the genetic resources can be effectively prevented from becoming extinct and bad influences and damage to the environment cannot be caused.
