The cloning method comprises the specific steps: A, determining an NtIPT1 gene sequence; B, extracting tobacco RNA and performing reverse transcription to obtain a first chain cDNA; C, designing and synthesizing a specific primer according to the NtIPT1 gene sequence and utilizing the cDNA as a template to perform PCR proliferation; D, recycling and purifying PCR products and constructing an overexpression carrier containing the NtNHX1-2 gene. The overexpression carrier is converted by agrobacterium mediated transformation to be expressedin tobacco to prepare transgenic seedlings; potassium content contrast of the obtained transgenic seedlings is improved by 20% or more, so that the tobacco NtNHX1-2 gene has an important applicationprospect in the aspect of cultivating tobacco with a high potassium content.
