The culture medium comprises, on the basis of 1000 ml, 10 to 30 ml of tobacco leaf juice, 20 to 32 ml of sweet white wine and 10 to 25 g of agar, and water is used as a blending agent to blend the culture medium to a rated amount; the pH value of the culture medium is 5.5 to 7.5. A formula for the culture medium is as follows: 10 to 30 ml of tobacco leaf juice, 20 to 32 ml of sweet white wine, 10 to 25 g of agar, 1000 ml of water and a pH value of 5.5 to 7.5. The preparation method comprises steps consisting of pre-treatment, preparation of a culture medium material, preparation of the culture medium and aftertreatment and comprises the following concrete steps: processing the components of the culture medium; uniformly mixing the tobacco leaf juice and the sweet white wine by using water or oat juice as the blending agent for blending; adding the agar and heating the agar until the agar is completely dissolved; carrying out split charging and sterilization so as to prepare a finished product of the culture medium for Alternaria alternate. Tests prove that the culture medium provided by the invention obviously accelerates the growth speed of mycelia of Alternaria alternate, improves output of spores and reduces bacterial pollution, and the preparation method is simple and convenient to operate and has low cost.
