The invention discloses a tobacco anabasine synthesis regulation gene NtERF91 and a cloning method and application thereof. The nucleotide sequence of the anabasine synthesis regulatory gene NtERF91 is shown as SEQ ID: No.1, and the coded amino acid sequence is shown as SEQ ID: No.2. The invention also discloses a cloning method of the anabasine synthesis regulation gene NtERF91. The method specifically comprises the following steps of: A, determining an NtERF91 gene sequence; B, extracting tobacco RNA, and performing reverse transcription to obtain a first strand cDNA; C, designing and synthesizing a specific primer according to the NtERF91 gene sequence, and carrying out PCR amplification by taking the cDNA as a template; D, recovering and purifying the PCR product; and E, constructing an over-expression vector containing the NtERF91 gene, over-expressing the over-expression vector in tobacco through agrobacterium-mediated transformation, and preparing transgenic plants. The resultsshowed that the contents of anabasine in the upper, middle and lower parts of the transgenic plants are increased by 1.5 times, 1.6 times and 1.3 times respectively compared with the control group, which indicates that the tobacco NtERF91 gene has a larger application prospect in the aspect of cultivating tobacco with high anabasine content.
