The preparation method comprises: taking 28-32 g of oatmeal, adding 500 mL of water, heating in boiling-water bath for 15 min, filtering by using three layers of gauze, taking the filtrate, adding 55-65 g of dry tobacco leaves, 8-12 g of soybean powder and 16-20 g of agar, and adding distilled water to have a total volume of 1000 mL, and performing split charging and sterilization to obtain the alternaria alternata medium. The alternaria alternata medium is cheap and easily-available in compositions, the preparation method is simple and easy to operate, and after the alternaria alternata medium is applied to inoculation of alternaria alternata spawns, the spore output is greatly improved.
