The disclosure also includes a method for making a mRNA therapeutic composition, comprising reconstituting a film comprising purified PMP lipids in the presence of an ionizable lipid to produce a LPMP comprising the ionizable lipid, and loading into the LPMPs with one or more polynucleotides encoding one or more tumor antigenic or immunogenic polypeptides.
