METHOD FOR OBTAINING AGAROSE FROM AHNFELTIA TOBUCHIENSIS USED AS THE BASIS OF NUTRIENT MEDIA

FIELD: microbiology.
SUBSTANCE: invention relates to the field of microbiology for use as the basis of nutrient media. The method is carried out as follows. A threefold extraction is carried out with a chemical compound of calcium, which is used as a 1.5% aqueous solution of calcium oxide at a temperature of 90-100°C. The duration of each stage of extraction is 1-2 hours. The extracts are separated at each stage and mixed. The obtained extract is purified and gelled. The formed algae gel is crushed, washed by infusion in distilled water and dried. The resulting agar is poured with distilled water to its concentration of 5-10% by weight, melted in a water bath, sequentially frozen and thawed. After that, gelling and the gel formed from agar is pressed at a pressure of 50-100 Pa for 6-8 hours. The pressed gel from agar is poured with hydrogen peroxide in an amount of 2% by weight and kept for 2 hours at a temperature of 30°C and pH 9. The resulting agarose is crushed and pressed at a pressure of 50-100 Pa for 6-8 hours. The coagel formed from agarose is crushed and dried to a constant weight at a pressure of 120-130 Pa.
EFFECT: invention reduces labor intensity by reducing the number and duration of stages, ensuring high efficiency of extraction of target products, increasing production safety due to the use of a more environmentally friendly reagent, expanding the scope, improving the quality of the finished product due to the low content of sulfates.
